VSG mRNA levels are regulated by the production of functional VSG protein

چکیده

The bloodstream form of Trypanosoma brucei persists in mammalian hosts through a population survival strategy depending on antigenic variation cell surface coat composed the variant glycoprotein (VSG). integrity VSG is essential and blocking its synthesis results division cycle arrest just prior to cytokinesis. This observation indicates that levels are monitored has mechanisms respond disruption synthesis. Here, regulation mRNA been investigated by first measuring copy number, second using ectopic expression transgenes containing premature termination codons. findings (i) number varies with identity (ii) pathway detects non-functional protein an increase levels.

mRNA levels Are Regulated by Muscle Activity

Denervation of adult skeletal muscle results in increased sensitivity to acetylcholine in extrajunctional regions of the muscle fiber. This increase in acetylcholine sensitivity is accompanied by a large increase in the level of mRNAs coding for the a-, f3-, Y-, and S-subunits of the acetylcholine receptor. To determine whether muscle activity is sufficient to regulate expression of extrajuncti...

Nucleosomes are depleted at the VSG expression site transcribed by RNA polymerase I in African trypanosomes.

In most eukaryotes, RNA polymerase I (Pol I) exclusively transcribes long arrays of identical rRNA genes (ribosomal DNA [rDNA]). African trypanosomes have the unique property of using Pol I to also transcribe the variant surface glycoprotein VSG genes. VSGs are important virulence factors because their switching allows trypanosomes to escape the host immune system, a mechanism known as antigeni...

Control of VSG gene expression sites.

Trypanosoma brucei survives in mammals by antigenic variation of its surface coat consisting of variant surface glycoprotein (VSG). Trypanosomes change coat mainly by replacing the transcribed VSG genes in an active telomeric expression site by a different VSG gene. There are about 20 different expression sites and trypanosomes can also change coat by switching the site that is active. This rev...

Cohesin regulates VSG monoallelic expression in trypanosomes

Antigenic variation allows Trypanosoma brucei to evade the host immune response by switching the expression of 1 out of approximately 15 telomeric variant surface glycoprotein (VSG) expression sites (ESs). VSG ES transcription is mediated by RNA polymerase I in a discrete nuclear site named the ES body (ESB). However, nothing is known about how the monoallelic VSG ES transcriptional state is ma...
